Research Associate, Cell Lines

Casma Therapeutics is harnessing the natural cellular process of autophagy to open vast new target areas for drug discovery and development that will profoundly impact the lives of patients. Autophagy is a conserved cellular process that contributes to overall organismal health. Inefficient autophagy flux or defects in lysosomal homeostasis are observed in numerous pathologies, e.g., neurodegeneration, metabolic disorders, inflammation, and muscle degeneration. Casma uses several approaches to intervene at strategic points throughout the autophagy-lysosome system to improve the cellular process of clearing out unwanted proteins, organelles and invading pathogens. By boosting autophagy, Casma expects to be able to arrest or reverse the progression of lysosomal storage disorders, muscle disorders, inflammatory disorders and neurodegeneration, among other indications. Casma was launched in 2018 by Third Rock Ventures and is based in Cambridge, Mass.
We are seeking a highly motivated, Research Associate to participate in drug discovery programs that lead to the identification of new targets and drug candidates for clinical development. The individual should possess strong core competencies in cell biology, molecular biology, assay development. We are seeking a qualified candidate who will share our excitement about the science, contribute to our culture, and honor our commitment to patients.
- Cell culture and cell line development using retro- and lenti-virus, CRISPR and downstream analysis of autophagy and signaling.
- Perform western blotting, FACS and imaging analysis to analyze autophagy and specific target activity in primary cells and cell lines.
- Aid in conducting genome-wide CRISPR screens for novel target ID.
- Validate hits from CRIPSR screens for autophagy regulators to aid target ID efforts.
- Use mouse tissue to both aid in the development of novel biomarkers and analyze key targets by protein and gene expression.
- Work to validate hits from high throughput screen using medium-throughput biochemical and high content cellular imaging assays.
- Develop tools to identify compound MOA.
- Work with primary neurons and patient derived cells to assess target engagement and disease efficacy.

Requirements + Qualifications
- Bachelor’s degree in biology or related discipline plus 3-5 years or Master’s degree plus 1-3 years of experience conducting high quality science in an industry setting
- Proficient in mammalian cell culture and cell line generation
- Experience with western blotting, immunoprecipitation, and plasmid preparation
- Expertise in genetic manipulation using CRISPR, shRNA, lentiviral/retroviral transduction or RNAi
- Hands-on knowledge of flow cytometry and FACS analysis techniques preferred
- Excellent organizational skills and attention to detail
- Strong interpersonal and collaboration skills
- Clear and effective verbal and written communication skills
- A demonstrated ability to think logically and critically
- Ability to successfully participate in a highly-creative and enthusiastic start-up environment
